Genetic bases for common polygenic diseases.
The present group of articles on the genetic basis of common diseases inaugurates a new policy on the part of the editors of the Proceedings. The editors plan to present a series of written minisymposia on dynamic and emerging areas of research of interest to broad groups of readers. We will solicit and present groups of interrelated articles by leading authorities and contributors describing t...
متن کامل[The genetic determinism of polygenic diseases].
Progress in molecular biology has opened the way to identifying genes involved in predisposition to multigene diseases. The two methods currently used for this purpose--analysis of candidate genes and systematic genomic screening--have given interesting but only very partial results. The problem is complicated by the large number of genes involved, their low penetrance, and linkage disequilibrium.
متن کاملA Polygenic Approach to the Study of Polygenic Diseases
Polygenic diseases are caused by the joint contribution of a number of independently acting or interacting polymorphic genes; the individual contribution of each gene may be small or even unnoticeable. The carriage of certain combinations of genes can determine the occurrence of clinically heterogeneous forms of the disease and treatment efficacy. This review describes the approaches used in a ...
متن کاملPolygenic score prediction captures nearly all common genetic risk for Alzheimer's disease
We estimate the maximum prediction accuracy for the risk of Alzheimer's disease based on disease prevalence and heritability of liability. We demonstrate that the recently reported AUC values for predicting of Alzheimer's disease using polygenic scores reach about 90% of the estimated maximum accuracy that can be achieved by predictors of genetic risk based on genomic profiles.
متن کاملGenetic evidence for common pathways in human age-related diseases
Aging is the single largest risk factor for chronic disease. Studies in model organisms have identified conserved pathways that modulate aging rate and the onset and progression of multiple age-related diseases, suggesting that common pathways of aging may influence age-related diseases in humans as well. To determine whether there is genetic evidence supporting the notion of common pathways un...
